Play whatever you feel like playing...
I wouldn't play NL50 HU with less than $1500 bankroll tho. I'd suggest HUSNGs if you don't have enough BR to play NL50 HU. They are more focused on push/fold and straightforward game than cash, but postflop play is still more advanced than in 6max games.
I think that HU will certainly improve your 6max skills also... I went from NL20 6max (had taken some shots at NL50 6max) to play NL20 HU (and so on up to NL100 HU) and now I've played some 6max again after half a year of HU and I'm up about 16 buy-ins at NL100 6max over a smallish sample.
So, I went from NL20 grinder (small winrate at NL50 6max over like 10k hand sample) to someone who feels completely confident playing NL100 6max, without actually playing 6max...
But anyway, if your only goal is to improve your 6max skills, then playings shorthanded is of course better. But HU experience will certainly improve your postflop game in any poker form... and most importantly, HU is fun!
